Il suo vero nome era Gladys Marie Smith.

Tra le più famose dive del cinema muto, costruì la sua immagine (e un'immensa fortuna finanziaria) grazie all'aspetto minuto fondato su uno sguardo di tenerezza e dai suoi biondi riccioli, che all'epoca rappresentarono un'autentica novità nella foggia della figura femminile; look ripreso in seguito da attrici come Shirley Temple e Judy Garland.

Figlia d'arte (sua madre, Charlotte Smith, attrice di teatro che esordì nel cinema praticamente negli stessi anni della figlia divenendone poi la sua manager), da adolescente, insieme ai fratelli Jack e Lottie, si misura con il teatro di provincia in Canada ma senza successo. Si trasferisce in America e a Broadway ha la possibilità di mettere in mostra il proprio talento. Il passo verso il cinema è brevissimo. Scoperta da D. W. Griffith, che la utilizzò in 25 film nel primo decennio del secolo, ha prolungato la carriera d'attrice in circa 250 lungometraggi con ben 184 apparizioni solo nei primi quattro anni, dal 1909 al 1912.

Poco prima degli anni '20 guadagna circa 12.000 dollari a settimana quando un bracciante ne percepisce 10.

Conosciuta con molti soprannomi ("la fidanzata d'America", "La piccola Mary", "L'enfant prodige", "Mary Blondielocks") ha lavorato da uno studios all'altro; dalla Biograph alla IMP, dalla Majestic alla Paramount, prima di fondare, nel 1919, insieme a Charlie Chaplin, D.W. Griffith e Douglas Fairbanks, la casa di produzione e distribuzione United Artists.

Cessa la sua attività d'attrice con l'avvento del sonoro, per dedicarsi esclusivamente alla produzione, dove investirà la sua già ingente fortuna quintuplicandone il valore. Si ritira definitivamente da ogni attività poco dopo gli anni '50, per godersi il suo immenso patrimonio.

Beverly Hills, il più ricco quartiere d'America, deve la sua popolarità al primo edifico costruito nel 1920 su un'area disabitata, il "Pickfair", principesca residenza dell'attrice e di suo marito Douglas Fairbanks (il nome deriva dalle iniziali dei due coniugi). Nel corso dei decenni successivi, attorno a questo palazzo principesco, venduto nel 1988 per 12 milioni di dollari, si allinearono le più fastose ville delle star hollywoodiane.

Per propria volontà testamentaria, circa 40 pellicole furono distrutte subito dopo i suoi funerali, poiché l'attrice le riteneva inadeguate, in quanto appariva "brutta".

Sposata tre volte con due divorzi e sempre con attori: Owen Moore, Douglas Fairbanks e Charles Rogers dal quale ha avuto due figli.

Hanno detto di lei - Samuel Goldwyn (produttore): "…ci vuole più tempo a redigere un contratto con Mary Pickford che girare i suoi film…"
