Studi e formazione: Autodidatta. Frequenta la scuola d'obbligo per soli tre mesi e apprende la lettura attraverso i cartelloni pubblicitari esposti per strada.
Tra le più famose dive del cinema muto, costruì la sua immagine (e un'immensa fortuna finanziaria) grazie all'aspetto minuto fondato su uno sguardo di tenerezza e dai suoi biondi riccioli, che all'epoca rappresentarono un'autentica novità nella foggia femminile; look ripreso in seguito da attrici come Shirley Temple e Judy Garland.
Figlia d'arte (sua madre, Charlotte Smith, attrice di teatro che esordì nel cinema praticamente negli stessi anni della figlia divenendone poi la sua manager), da adolescente, insieme ai fratelli Jack e Lottie, si misura con il teatro di provincia in Canada ma senza successo, all'infuori della rappresentazione del ´Bootle's Baby´ del 1898, dove si esibisce in tuor attraverso la compagnia Valentine Stock Company. Si trasferisce in America e nel 1907 esordisce a Broadway nel ´The Warrens of Virginia´.
Il passo verso il cinema è brevissimo. Due anni dopo è scoperta da D. W. Griffith - che la utilizzò in 25 film nel primo decennio del secolo - inizia ad essere interprete per film di corto e medio metraggio e, in seguito, in circa 250 lungometraggi con ben 184 apparizioni solo nei primi quattro anni, dal 1909 al 1912.
Poco prima degli anni '20 guadagna circa 12.000 dollari a settimana quando un bracciante ne percepisce 10.
Conosciuta con molti soprannomi ("La fidanzata d'America", "La piccola Mary", "L'enfant prodige", "Mary Blondielocks") ha lavorato da uno studios all'altro; dalla Biograph alla IMP, dalla Majestic alla Paramount e uno suo di proprietà, la Mary Pickford Productions, prima di fondare, nel 1919, insieme a Charlie Chaplin, D.W. Griffith e Douglas Fairbanks, la casa di produzione e distribuzione United Artists.
Cessa la sua attività d'attrice pochi anni dopo l'avvento del sonoro, per dedicarsi esclusivamente alla produzione, dove investe la sua già ingente fortuna quintuplicandone il valore. Si ritira definitivamente da ogni attività poco dopo gli anni '50, per godersi il suo immenso patrimonio.
Nel 1950, Billy Wilder la richiama per interpretare Norma Desmond nel noir Viale del tramonto, ma per strane situazioni che si accavallarono tra loro, la parte andò ad un altra diva del muto ormai ferma da circa un decennio, Gloria Swanson.
Nell'aprile del 1927, è stata la priva diva, insieme al marito Douglas Fairbanks, a inauguare ufficialmente "l'impronta della mano" nel cemento davanti al Grauman's Chinese Theatre.
Beverly Hills, il più ricco quartiere d'America, deve la sua popolarità al primo edifico costruito nel 1920 su un'area disabitata, il ´Pickfair´, principesca residenza dell'attrice e di suo marito Douglas Fairbanks (il nome deriva dalle iniziali dei due coniugi). Nel corso dei decenni successivi, attorno a questo palazzo principesco, venduto nel 1988 per 12 milioni di dollari, si allinearono le più fastose ville delle star hollywoodiane.
Per propria volontà testamentaria, circa 40 pellicole furono distrutte subito dopo i suoi funerali, poichè l'attrice le riteneva inadeguate, in quanto, a suo dire, appariva brutta e, soprattutto, per paura che nessuno se ne sarebbe mai preso cura.
Muore a 86 anni per emorragia cerebrale.
Sposata tre volte con due divorzi e sempre con attori: Owen Moore, Douglas Fairbanks e Charles Rogers (due figli adottati, Ronald Charles e Roxanne, morta nel 2007 per osteoporosi).
Tra i matrimoni, è stata amante degli attori Leslie Howard, Thomas Meighan, Kenneth MacKenna, Marshall Neilan, Craig Reynolds e dei registi Frank Borzage e William Desmond Taylor.
